- Pembrolizumab plus axitinib versus sunitinib for advanced clear cell renal cell carcinoma: 5-year survival and biomarker analyses of the phase 3 KEYNOTE-426 trial
Nature Medicine, Published online: 01 August 2025; doi:10.1038/s41591-025-03867-5Five-year follow-up and prespecified exploratory biomarker analysis from the phase 3 KEYNOTE-426 trial show that pembrolizumab plus axitinib compared to sunitinib as first-line therapy continues to show overall and progression-free survival benefits in patients with advanced clear cell renal cell carcinoma and indicate that a T-cell-inflamed gene expression profile is predictive of response.
- Intravenous iron therapy reduces cardiovascular events in heart failure
Nature Medicine, Published online: 01 August 2025; doi:10.1038/s41591-025-03862-wIntravenous iron treatment lowers the risk of repeated hospitalizations for heart failure and cardiovascular death in patients with iron deficiency and heart failure with reduced left ventricular ejection fraction. The therapy has a good safety profile, and its benefits are most pronounced within the first year of treatment, when iron dosing is highest.

- Multimodal AI correlates of glucose spikes in people with normal glucose regulation, pre-diabetes and type 2 diabetes
Nature Medicine, Published online: 31 July 2025; doi:10.1038/s41591-025-03849-7Multimodal data from 347 deeply phenotyped individuals including healthy, prediabetic individuals and individuals with T2D report remotely acquired patterns of glucose control via continuous glucose monitoring, and correlates them with diet and microbiome features and physiological signals, showing that these are able to discriminate individuals with T2D from control also in a large independent cohort.
- Permissive gun laws in the USA linked to thousands of child deaths
Nature Medicine, Published online: 31 July 2025; doi:10.1038/d41591-025-00048-2A new analysis shows that more-permissive state-level firearm laws have contributed to thousands of excess firearm-related child deaths, while a few states enacting stricter laws have seen these death rates decrease.
- A HER3-targeting antibody–drug conjugate for leptomeningeal metastasis
Nature Medicine, Published online: 31 July 2025; doi:10.1038/s41591-025-03853-xThe phase 2 TUXEDO-3 trial identifies the HER3-targeting antibody–drug conjugate patritumab deruxtecan as a promising new therapeutic option for patients with leptomeningeal metastasis from solid tumors.
